Efficiency of Reshaping Hydroxyl Protection Reaction with Pyridine p-Toluene Sulfonate

Against the backdrop of the continuous rise in innovative drug research and development in the industry, pyridinium p-toluenesulfonate (PPTS) is becoming a popular “tool” in the field of organic synthesis due to its unique catalytic properties. As an ideal alternative to p-toluenesulfonic acid (PTSA), this white crystalline powder exhibits irreplaceable process advantages in hydroxyl protection and deprotection reactions.

The core value of PPTS lies in its “mild and efficient” catalytic properties. Although traditional p-toluenesulfonic acid has strong catalytic activity, it exists in the form of monohydrate crystals with strong acidity, which can easily damage acid sensitive functional groups in the molecule. In contrast, PPTS is an anhydrous compound with milder acidity, capable of precise functional group conversion in complex molecular frameworks. Researchers can sequentially carry out desilication and etherification reactions of hydroxyl groups and intramolecular acetalization reactions in the same reaction system, thereby efficiently constructing heterocyclic compounds or lactone structures. The characteristics of simple operation and mild conditions greatly simplify the synthesis route and improve the yield of the target product. Currently, with the continuous development of GLP-1 peptide drugs and antibody conjugated drugs (ADCs), the demand for highly selective and compatible catalytic reagents is becoming increasingly urgent. PPTS provides full cycle support from laboratory research and development to pilot scale for innovative pharmaceutical companies with its stable supply capacity of over 120 tons per year.
